چکیده

PurposeCancer stem cells constitute an endless reserve for the maintenance and progression of tumors, they could be reason conventional therapy failure. New therapeutic strategies are necessary to specifically target them. In this context, microsecond pulsed electric fields have been selected expose D283Med cells, a human medulloblastoma cell line resulted rich in cancer normal astrocytes.MethodsWe analyzed vitro different endpoints at times after field exposure, such as permeabilization, reactive oxygen species generation, viability/proliferation, cycle, clonogenicity, well expression genes involved apoptosis, senescence. Furthermore, response exposed was validated vivo heterotopic mouse xenograft model.ResultsOur results showed that specific pulse protocol (ie, 0.3 MV/m, 40 μs, 5 pulses) able induce irreversible membrane permeabilization apoptosis exclusively cells. surviving generation observed, together with transitory G2/M cell-cycle arrest senescence-associated phenotype via upregulation GADD45A. results, demonstrated significant tumor volume reduction no eradication mass. conjunction, we verified efficacy pre-exposure followed by ionizing irradiation enable complete inhibition growth.ConclusionsOur data reveal novel options targeting indicating nonionizing effective means overcome radioresistance Cancer astrocytes. We model. Our growth.

Inhibition of PI3K Signalling Selectively Affects Medulloblastoma Cancer Stem Cells

Medulloblastoma is the most common malignant brain tumor of childhood. Although survival has slowly increased in the past years, the prognosis of these patients remains unfavourable. In this context, it has been recently shown that the intracellular signaling pathways activated during embryonic cerebellar development are deregulated in MDB. One of the most important is PI3K/AKT/mTOR, implicated...

RAD51 Is a Selective DNA Repair Target to Radiosensitize Glioma Stem Cells

Patients with glioblastoma die from local relapse despite surgery and high-dose radiotherapy. Resistance to radiotherapy is thought to be due to efficient DNA double-strand break (DSB) repair in stem-like cells able to survive DNA damage and repopulate the tumor. We used clinical samples and patient-derived glioblastoma stem cells (GSCs) to confirm that the DSB repair protein RAD51 is highly ex...

Cryosurgery with Pulsed Electric Fields

This study explores the hypothesis that combining the minimally invasive surgical techniques of cryosurgery and pulsed electric fields will eliminate some of the major disadvantages of these techniques while retaining their advantages. Cryosurgery, tissue ablation by freezing, is a well-established minimally invasive surgical technique. One disadvantage of cryosurgery concerns the mechanism of ...

New Molecules and Old Drugs as Emerging Approaches to Selectively Target Human Glioblastoma Cancer Stem Cells

Despite relevant progress obtained by multimodal treatment, glioblastoma (GBM), the most aggressive primary brain tumor, is still incurable. The most encouraging advancement of GBM drug research derives from the identification of cancer stem cells (CSCs), since these cells appear to represent the determinants of resistance to current standard therapies. The goal of most ongoing studies is to id...
